Characterization of different G protein coupling properties of CB1 and CB2 cannabinoid receptors and GPR55 receptor using BRET
نویسندگان
چکیده
Background CB1 and CB2 cannabinoid receptors are G protein-coupled receptors which have been described to couple mainly to the Gi/o subfamily of G proteins. However, in some cell types and upon stimulation with certain cannabinoid agonists, activation of other G protein subtypes has also been observed. GPR55 is an orphan G protein-coupled receptor which has been suggested to be a novel member of the cannabinoid receptor family.

Functional characterization of the novel cannabinoid receptor GPR55 and its modulation by CB1 and CB2 receptors
We have recently shown that the G protein-coupled receptor 55 (GPR55) mediates intracellular effects of cannabinoids and other, non-cannabionid, ligands in addition to the classical cannabinoid 1 (CB1R) and 2 (CB2R) receptors. Here we show that the presence of either CB1R or CB2R alters the signaling pathways of GPR55. HEK-293 cells stably expressing either the GPR55 receptor alone or in combin...
متن کاملThe orphan receptor GPR55 is a novel cannabinoid receptor.
BACKGROUND The endocannabinoid system functions through two well characterized receptor systems, the CB1 and CB2 receptors. Work by a number of groups in recent years has provided evidence that the system is more complicated and additional receptor types should exist to explain ligand activity in a number of physiological processes. EXPERIMENTAL APPROACH Cells transfected with the human cDNA ...
متن کاملRod Photoreceptors Express GPR55 in the Adult Vervet Monkey Retina
Cannabinoids exert their actions mainly through two receptors, the cannabinoid CB1 receptor (CB1R) and cannabinoid CB2 receptor (CB2R). In recent years, the G-protein coupled receptor 55 (GPR55) was suggested as a cannabinoid receptor based on its activation by anandamide and tetrahydrocannabinol. Yet, its formal classification is still a matter of debate. CB1R and CB2R expression patterns are ...
متن کاملGPR55: signaling pathways and functions
Background We have recently shown that the G protein-coupled receptor 55 (GPR55) mediates intracellular effects of cannabinoids and other, non-cannabinoid ligands in addition to the classical cannabinoid 1 (CB1) and 2 (CB2) receptors. Here we show different signaling pathways triggered by GPR55 in response to a panel of its agonists. In addition the cytoskeleton rearrangement mediated by GPR55 ...
متن کاملEffect of Cannabinoid Receptor Activation on Spreading Depression
Objective(s) The objective of this study was to evaluate the effect of cannabinoid on cortical spreading depression (CSD) in rat brain. Cannabis has been used for centuries for both symptomatic and prophylactic treatment of different types of headaches including migraine. CSD is believed to be a putative neuronal mechanism underlying migraine aura and subsequent pain. Materials and Methods T...
